Plateforme de Gestion des Echantillons Biologiques – PGEB
La PGEB
La Plateforme de Gestion des Echantillons Biologiques (PGEB) a été créée en 2010 afin de regrouper les moyens techniques et humains au service de la recherche fondamentale, translationnelle et clinique.
Elle regroupe le Centre de Ressources Biologiques (CRB) créé en 2004, qui intègre la tumorothèque historique et la sérothèque, le Bureau des Essais Cliniques (BEC) et la plateforme d’extraction d’acides nucléiques.
Nos missions
La plateforme assure au quotidien la gestion des ressources biologiques dans le cadre de :
- Diagnostic : conservation de tissus congelés
- Centre de Ressources Biologiques (CRB): collection institutionnelle et essais cliniques de promus par le CLB
- Mise à disposition de tissus frais : avec l’aide de la Plateforme Ex-Vivo
- Essais cliniques de promotion externe
- Préparation de produits dérivés (ADN,ARN)
- Hébergement de collections externes : UNICANCER, MESOBANK, SHARE4Kids

Collections et mise à disposition des ressources biologiques
La collection institutionnelle du Centre Léon Bérard a été constituée dans le cadre de la recherche et du diagnostic en cancérologie regroupant différents types de pathologies:
Tumeurs du sein, sarcomes, tumeurs de l’ovaire, tumeurs du poumon …
Elle intègre une large variété de types d’échantillons : tissus, fluides biologiques (sang, selles, salive, …), cellules (PBMC) et acides nucléiques.
- Plus de 8000 échantillons en Tumorothèque
- Plus de 45000 échantillons en Sérothèque : Sang, plasma, sérum, cellules PBMC, Buffy Coat, …
- Plus de 22000 échantillons de produits dérivés : ADN, ARN,…
La sérothèque contient les fluides biologiques issus des prélèvements des patients du centre et des préparations réalisés sur ces prélèvements :
- Sang total : 5000 échantillons
- Plasma : 20 000 échantillons
- Sérum : 11 000 échantillons
- Cellules : 8000 échantillons
- Buffy Coat, liquide pleurale, urine, …
La PGEB réalise, par une expertise scientifique et des moyens
techniques avancés et précis, des extractions d’acides nucléiques à partir de:
- Tissus : congelés, inclus en paraffine, poudres, …
- Fluides biologiques : Sang, salive, …
- Cellules congelées, frottis buccaux, …
La collection de produits dérivés contient plus de 14000 échantillons d’ADN et plus de 8000 échantillons d’ARN
Le CLB est aussi le centre hébergeur de la collection biologique de la fédération UNICANCER. Dans ce cadre, la PGEB assure la réception, la conservation et la mise à disposition des échantillons de cette collection.

Structuration
Une équipe de 18 personnes sous la responsabilité scientifique de Séverine TABONE-EGLINGER, assure quotidiennement la gestion des échantillons biologiques destinés à la communauté médico-scientifique du centre, ainsi qu’à ses partenaires académiques et industriels.
Elle est composée de :
– une chef d’équipe (Séverine MARTINEZ)
– une coordinatrice des essais cliniques (Magali BONIFACIO)
– 13 techniciens et 2 aides-laboratoire
Expertises
– Gestion d’échantillons dans le cadre d’essais cliniques.
– Préparation de matériel sanguin (sérum, plasma, plasma CtDNA…)
– Purification de cellules mononuclées – PBMC (sang ou moelle), tri magnétique CD34+
– Extractions de produits dérivés (ADN, ARN) à partir de tout type d’échantillons
– Contrôles qualité sur les échantillons : viabilité de cellules, pourcentage de cellules tumorales, qualité ADN/ARN, stabilité ADN…
– Conservation d’échantillons (azote, -80°C ou ambiant contrôlé) dans des conditions maîtrisées et tracées
– Mise à disposition d’échantillons de qualité au niveau national et international (IATA)
Réseaux et démarche Qualité
La PGEB veille à ce que ses activités soient conformes aux normes du biobanking en vigueur, elle est donc certifiée selon les normes AFNOR NF S96-900 (depuis 2009) et ISO 9001 (depuis 2013).
Elle fait partie de plusieurs réseaux de biobanking au niveau national et international :
